摘要
目的研究C型产气荚膜梭菌α-β2-β1融合基因重组工程菌BL21(DE3)/pXETAB2B1抗原的免疫原性。方法在对重组工程菌BL21(DE3)/pXETAB2B1进行优化表达的基础上,制备包涵体粗提物、全细胞培养物及全细胞乳糖诱导培养物3种抗原,对ICR小鼠进行攻毒保护试验。结果重组工程菌BL21(DE3)/pXETAB2B1能够表达α-β2-β1融合蛋白,该融合蛋白能够有效刺激免疫小鼠产生特异性抗体,达到保护的目的。结论已获得具有一定免疫原性的重组工程菌,可作为预防产气荚膜梭菌所引起疾病的候选菌株。
Objective To study the immunogenicity of antigen of recombinant E. coli BL21 (DE3)/pXErAB2BI for expression of Clostridium perfringens α-β2-β1 fusion gene. Methods Based on the optimal expression of recombinant E. coli BL21 (DE3)/pXErAB2B1, 3 kind of antigens, i. e. crude extract of inclusion body, whole cell culture and lactose-induced whole cell culture, were prepared for immunization of ICR mice. Challenge the mice with virulent strain C59-2 of Clostridium peofringens and observe the survival. Results α-β2-β1 fusion protein was expressed in recombinant E. coli BL21 (DE3)/pXErAB2BI and induced specific antibody which protected the immunized mice against challenge with virulent strain of Clostridium perfringens. Conclusion Recombinant E. coli BL21 (DE3)/pXETAB2BI showed a certain immunogenicity and might be used as a candidate strain for prevention of diseases caused by Clostridium pefringens.
