光动力联合5-FU缓释剂治疗兔VX-2口腔鳞瘤 被引量:2

The study of photodynamic therapy with sustained-release 5-fluorouracil for the treatment of Vx-2 oral squamous cell carcinoma in rabbits

摘要 目的研究光动力学疗法(PDT)联合5-氟尿嘧啶(5-FU)在治疗口腔鳞癌中的应用。方法通过建立100只VX-2口腔鳞癌动物模型,随机分为5组。通过肿瘤体积测量、组织学观察和免疫组化等方法评价肿瘤抑瘤率、生长状态、p53蛋白表达情况来反应PDT联合5-FU的疗效,各组数据比较用SPSS11.0统计软件进行分析。结果治疗后第5周观察:①各组抑瘤率分别为静脉给药组39.9%±15.9%;5-Fu缓释剂组73.8%±22.3%;PDT组50.6%±21.8%;联合治疗组93.6%±19.5%,联合治疗组抑瘤率低于其他各组,差异具有显著性(P<0.05)。②HE染色镜下观察联合治疗组较其他组肿瘤细胞增殖能力下降,核分裂像罕见,细胞大量死亡或凋亡。③免疫组化测得联合治疗组p53阳性表达率为15%,与其他组比较差异具有显著性(P<0.05)。结论在口腔鳞癌治疗上PDT与5-FU缓释剂联合应用较其单独应用疗效显著,可能成为治疗口腔鳞癌的一种方法。 Objective To study the effect of photodynamic therapy(PDT) with 5-FU on the treatment of oral squamous cell carcinoma(OSCC). Methods One hundred rabbits were divided into five groups, which were implanted with VX-2 squamous cell carcinoma tissue pieces. Tumor growth, inhibition rate (IR) based on tumor measurement, histopathologic change, the expression of p53 protein were observed and recorded. The data was analyzed by SPSSll. 0 software package. Results The IR in PDT with 5-FU group (Group E) was significantly higher than that in other groups(P 〈0. 05 ). Histopathologically, the proliferation of the tumor cells was relatively suppressed in group E. The expression of p53 was showed that the lethal effect of group E was more obvious than the other groups. Conclusion Our study demonstrates that the combination of PDT and sustained-release 5-FU is more effective in OSCC.
