摘要
目的观察吡格列酮和白细胞介素-6(IL-6)对3T3-L1脂肪细胞Visfatin mRNA表达的影响。方法体外培养3T3-L1前体脂肪细胞并诱导其分化成熟,分别以不同浓度的吡格列酮和(或)IL-6干预,在不同的时间段提取细胞总RNA,采用RT-PCR技术检测细胞中Visfatin mRNA的表达水平。结果IL-6和吡格列酮分别作用于分化成熟的3T3-L1脂肪细胞均能下调Visfatin mRNA的表达,且用吡格列酮预处理或与IL-6共同作用均不能够改善IL-6对Visfatin mRNA的抑制作用。结论IL-6对Visfatin mRNA表达的负调控可能是其引起胰岛素抵抗和糖脂代谢紊乱的机制之一,Visfatin可能并不是噻唑烷二酮(TZD)类药物改善胰岛素敏感性的靶基因。
Objective To observe the effects of pioglitazone and interleukin-6 on visfatin mRNA expression in 3T3- L1 adipocytes. Methods 3T3-L1 preadipocytes were cultured in vitro and differentiated into the matured adipocytes. Pioglitazone and/or IL-6 with different concentrations were added into the culture medium of differentiated adipocytes for various time. Total RNA was extracted and visfatin mRNA expression was measured by semiquantitative RT-PCR technique. Results Pioglitazone and IL-6 both suppressed the visfatin mRNA expression in 3T3-L1 adipocytes. In our study, PPARγ/activation by pioglitazone was not sufficient to reverse IL-6-induced visfatin downregulation. Conclusion of the insulin resistance Downregulation of visfatin mRNA expression by IL-6 might play a role in the pathogenesis syndrome. Visfatin is not a target for TZD and, thus, and unrelated to the insulin-sensitizing effect of these drugs.
