摘要
目的研究干扰素α抑制乙肝病毒X蛋白诱导肝细胞癌侵袭转移的分子机制,为肝癌的临床治疗研究提供实验依据。方法采用脂质体法构建pcDNA3.1-HBx质粒转染CCL13细胞,将CCL13/HBx细胞与干扰素-α共培养,并用RT—PCR、Western Blot从mRNA和蛋白质水平检测CCL13/pcDNA3.1、CCL13/HBx、CCL13/HBx—INF—α中p202、PTEN基因表达,Transwell实验检测CCL13/pcDNA3.1、CCL13/HBx、CCL13/HBx—INF—α细胞侵袭能力。结果CCL13/HBx细胞中p202、PTEN基因的mRNA及蛋白表达水平明显低于CCL13/pcDNA3.1细胞和CCL13/HBx—INF—α细胞(P〈0.05),而CCL13/pcDNA3.1细胞与CCL13/HBx—INF—α细胞间表达差异无统计学意义(P〉0.05)。CCL13/HBx细胞通过Matrigel的细胞数[(37.2±3.8)个/HP]明显多于CCL13/pcDNA3.1[(6.4±1.2)个/HP,t=8.369,P〈0.05]和CCL13/HBx—INF—α细胞[(7.6±1.3)个/HP,t=7.256,P〈0.051,而CCL13/pcDNA3.1与CCL13/HBx—INF—α细胞间差异无统计学意义(P〉0.05)。结论干扰素α可能通过上调乙肝病毒X蛋白所介导的p202基因、PTEN基因表达,抑制肝癌细胞的侵袭转移。
Objective To investigate the molecular mechanism by which interferon-α inhibits the invasion and metastasis of hepatocellular carcinoma activated by hepatitis B virus X protein, and to found an experimental basis for clinical treatment of hepatocellular carcinoma. Methods pcDNA3.1-HBx plasmid was transfected into CCL13 cells by lipofectamine in vitro, CCL13/HBx was co-cultured with interferon-α. RT-PCR and Western blot was used to detect the expression of P202,PTEN in CCL13/pcDNA3.1 ,CCL13/ HBx and CCL13/HBx- INF-α at mRNA and protein level. Transwell test was used to detect the invasion ability of CCL13/pcDNA3. 1 ,CCL13/HBx,CCL13/HBx-INF-α. Results The expression of p202,PTEN in CCL13/HBx was significantly lower than that in CCL13/ pcDNA3. 1 and CCL13/ HBx-INF-α both at mRNA and protein level, hut there was no significant difference between CCL13/pcDNA3.1 and CCL13/ HBx-INF-α. More CCL13/ HBx cells passed through matrigel (37. 2 ± 3.8/HP) than CCL13/ pcDNA3. 1 (6. 4 ± 1.2/HP, t = 8. 369, P 〈 0. 05 ) and CCL13/HBx-INF-α (7. 6 ± 1.3/HP, t = 7. 256, P 〈 0. 05 ), but there was no difference between CCL13/ pcDNA3. 1 and CCL13/ HBx-INF-α (P 〉 0.05 ). Conclusion Interferon-α may inhibit the metastasis of hepatocellular carcinoma by up-regulating the expression of p202 and PTEN induced by Hepatitis B virus X protein.
