摘要
目的:观察心肌内源性硫化氢生成的变化及外源性给予硫化氢供体NaHS对心肌损伤的影响,以探讨硫化氢在心力衰竭中的病理生理意义。方法:结扎冠状动脉前降支诱导大鼠心力衰竭,腹腔给予外源性H2S治疗;HE染色检测心肌损伤程度;亚甲基蓝法测定硫化氢浓度;TBA法测定血浆和心肌脂质过氧化产物丙二醛(MDA)含量;自动生化分析仪检测血清天门冬氨酸氨基转换酶(AST)、磷酸肌酸激酶(CPK)及乳酸脱氢酶(LDH)活性。结果:大鼠冠状动脉结扎导致严重心肌损伤;血浆和心肌组织H2S生成显著减少(P<0.001);血浆和心肌MDA含量明显增加(P<0.05)。血浆AST、CPK和LDH活性显著增加(P<0.05)。给予NaHS治疗后,心肌损伤程度明显减轻;血浆和心肌MDA含量明显降低(P<0.05);血浆AST、CPK和LDH活性显著降低(P<0.05)。结论:H2S可通过减少血浆和心肌MDA的生成而对大鼠心肌缺血导致的心力衰竭具有保护作用。
Objective To investigate the changes of endogenous hydrogen sulfide (H-S) in rat with heart failure (HF) and the effects of exogenous administration of H-S donor sodium hydrosulphide (NariS) on myocardial infarction, so as to explore the pathophysiological significance of H-S in HF. Methods: The experimental HF model was induced by left anterior descending coronary occlusion and then was treated with intraperitoneal injection of H2S; the degree of myocardial infarction was determined by observing hematoxylin-eosin stained slices; the H2S level was measured by methylene blue; the contents of malondialdehyde (MDA) in plasma and myocardial homogenates were detected by TBA; aspartate amino transferase (AST), creatine phosphokinase (CPK) and lactate dehydrogenase (LDH) activities in serum were determined by automatic biochemistrical analysis apparatus. Results: Severe myocardial infarction was induced by coronary occlusion; H2S production in plasma and heart homogenates were significandy declined (P〈0.001); the content of MDA in plasma and heart homogenates was inereased (P〈0.05); AST, CPK and LDH activities in plasma were increased (each P〈0.05). After treatment with NariS, myocardial infarction was obviously rduced; the contents of MDA in plasma and heart homogenates were markedly declined (each P〈0.05); AST, CPK and LDH activities in plasma were decreased (each P〈 0.05). Conclusion: The results suggest that H2S has protective effects in HF following myocardial isehemia in rats by decreasing MDA production in plasma and heart.
