摘要
目的观察葛根素对微循环、脑缺血缺氧的改善作用及对垂体后叶素(Pit)所致大鼠急性心肌缺血的保护作用。方法用PeriFlux 5000激光多普勒探测小鼠静脉给药前和静脉给药后2min^5min耳廓微循环灌注量的变化。皮下注射给药6d,末次给药后30min,断头观察断头后喘息持续的时间。尾静脉注射垂体后叶素造成急性心肌缺血模型,观察各给药组在不同时间点Ⅱ导联心电图心率、ST段变化及伴心律失常等心电图变化。给药1h后眼眶静脉丛采血分离血清,测血清天门冬酸氨基转移酶(AST)、肌酸激酶(CK)、乳酸脱氢酶(LDH)。结果葛根素给药组与模型组比较,小鼠耳廓微循环灌注量明显增加,断头后喘息时间延长。葛根素也能明显对抗垂体后叶素引起的急性心肌缺血的ST段抬高,降低心律失常的发生率,及降低血清AST、CK、LDH。结论葛根素具有改善微循环及脑缺血缺氧的作用,对垂体后叶素所致大鼠急性心肌缺血具有保护作用。
Objective To observe the effect of puerarin on microcirculation and acute myocardial ischemia. Methods Blood perfusion of auris microcirculation was detected before and after injection of puerarin within 2 - 5 minutes with PeriFlux 5000 laser Doppler blood equipment. Acute myocardial ischemia models were established by injection of pitui.tary. The electrocardiogram and serum contents of lactate dehydrogenase (LDH), creatine kinase (CK), and aspartate transaminase (AST) were observed after pituitary injection. Results To compare with the model group, the blood perfusion of auris microcirculation in puerarin group improved and the breathing time was prolonged. The levels of CK, LDH and AST in puerain group deceased. Conclusion Puerarin could improve microcirculation, decrease oxygen consumption and improve acute myocardium isehemia.
