α-硫辛酸对小鼠急性胰腺炎NF-κB活性影响的实验研究

摘要 目的观察α-硫辛酸对小鼠急性胰腺炎NF-κB活性的影响,探讨其对急性胰腺炎的保护作用及机制。方法选用健康昆明种小鼠96只,随机分为对照组、模型组和干预组。采用腹腔内注射L-精氨酸法建立小鼠急性胰腺炎模型,于术后第6、12、24、48小时分批处死小鼠,取胰腺组织进行病理评分,并测定胰SOD、MDA活性;免疫组化法检测胰腺细胞NF-κB活化情况。结果α-硫辛酸干预治疗后NF-κB活化被抑制(P<0.05);SOD上升,MDA降低(P<0.05);胰腺病理评分也降低(P<0.05)。结论α-硫辛酸可抑制急性胰腺炎时胰腺细胞NF-κB活化,其机制可能是硫辛酸能改善胰腺氧化应激状态,从而减轻胰腺的病理损害。 Objective To observe the effect of alpha-lipoic acid (LA)on the levels of NF-κB signal pathway in pancreatic tissue with acute pancreatitis(AP)and to study the protective mechanism of LA on acute pancreatitis. Methods All the mice were randomly divided into three groups:normal control group; AP group-acute pancreatitis group; LA group. Mice were injected with L-arginine for 2 times with an interval of 1 h. An hour before, LA was injected intramuscularly. At 6 h, 12 h, 24 h and 48h after injection, the animals were sacrificed. The pancreas were removed for histological observation and the activity of NF-κB in pancreatic tissue was measuredby immunohistochemistry. Results Histologically, LA reduced the severity of pancreatic injury including inflammatory cell infiltration and necrosis. The level of MDA in LA group was lower than that in the AP group(P〈0. 05),the level of SOD was higher in LA group than that in the AP group(P〈0.05). The activity of NF-κB decreased in LA pretreated group than those in AP group (P〈0.05). Conclusion The effects of LA as an antioxidant might be related to the inhibition of NF-κB activity. The results suggest that LA would prove to be useful for the treatment of AP.
