摘要
目的探讨凋亡抑制Survivin蛋白在人脑星形细胞瘤组织中的表达及其与脑星形细胞瘤发生、发展的关系。方法应用免疫组织化学Elivision二步法检测67例脑星形细胞瘤组织和10例对照组组织标本中Survivin蛋白的表达水平。结果对照组阳性表达率明显低于脑星形细胞瘤Ⅰ~Ⅳ级组(P〈0.01)。高分化组(脑星形细胞瘤Ⅰ~Ⅱ级组)阳性表达率为24.24%,明显低于低分化组(脑星形细胞瘤Ⅲ~Ⅳ级组)的70.59%(P〈0.01)。相关性分析结果显示,肿瘤分化高低与Survivin蛋白的表达强度呈正相关(r=0.437,P〈0.01)。结论Survivin蛋白在脑星形细胞瘤组织中呈高表达;且随脑星形细胞瘤病理级别的增高,阳性表达率和表达强度增强。Survivin蛋白在脑星形细胞瘤发生、发展过程中具有重要作用。
Objective To examine the expression level of Survivin in the human astrocytomas and its relationship between tumorigenesis and progression of the human astrocytomas. Methods Expression level of Survivin protein was investigated by immunohistochemistry Eiivision method in 67 cases(grade Ⅰ 16 cases,grade Ⅱ 17 cases,grade Ⅲ 17 cases,grade Ⅳ 17 cases)of the human astrocytomas and 10 cases of normal brain tissue. Results The positive percentage of Survivin in 10 cases of normal brain tissue was lower Significantly than that in the human astrocytomas(from Ⅰ to Ⅳ grade) (P〈0.01). The positive percentage of lower grade( Ⅰ~ Ⅱ )was 24.24% ,which was lower Significantly than those of higher grade (Ⅲ~Ⅳ ) that was 70.59% (P〈0.01). Spearman's Correlation Coefficient, along with the pathological grade increasing of human astrocytomas,the positive percentage and expressive intensity of Survivin increased gradually(r=0. 437 , P〈0. 01). Conclusion Survivin is overexpressed in the human astrocytomas,and the positive percentage and expressive intensity of Survivin is improved with the increasing of pathological grade of the human astrocytomas. Survivin may play an important role in tumorigenesis and progression of the human astrocytomas.
