摘要
目的:探讨贝那普利和厄贝沙坦联用对心衰大鼠血管内皮功能的影响。方法:SD大鼠45只,采用缩窄大鼠腹主动脉造成压力负荷性心力衰竭的大鼠模型。分为5组:假手术组、模型组、贝那普利组(10mg/kg·d)、厄贝沙坦组(50mg/kg·d)及联合用药组(Ben 5mg/kg·d+Irb 25mg/kg·d),连续给药8周。检测血清超氧化物歧化酶(SOD)活性,丙二醛(MDA)、一氧化氮(NO)、血浆AngⅡ和ET的含量。结果:模型组NO和SOD显著降低(P<0.01),MDA、ET和AngⅡ显著升高(P<0.01),治疗后各组NO和SOD显著升高(P<0.01),MDA和ET显著降低(P<0.01),其中联合用药组MDA低于贝那普利组(P<0.05),SOD高于厄贝沙坦组(P<0.05);治疗后,贝那普利组AngⅡ水平明显降低(P<0.01),而厄贝沙坦组明显升高(P<0.01),联合用药组明显低于模型组和单用组(P<0.01)。结论:联合应用贝那普利和厄贝沙坦对改善心衰大鼠血管内皮功能具有协同作用。
Objective:To investigate the action of Benazepril,Irbesartan and the union of the two on blood vessel endothelial injury induced by oxidative stress of CHF rats.Methods:The cardiac failure animal model in 45 SD rats were made by partially banding abdominal aorta to get pressure load.The animals were divided into five groups: the heart failure group ,the control group , the Benazepril group (10mg/kg/d), the Irbesartan group (50mg/kg/d) and the combination group (Ben 5mg/kg/d+Irb 25mg/kg/d ), with drugs treated for 8 weeks.The serum contents of SOD , MDA and NO were detected ; The plasma Ang Ⅱ and ET were determined by radioimmunoassay. Results:NO and SOD of Mod significantly decreased compared with Con(P〈0.01). MDA, ET and Ang Ⅱ increased (P〈 0.01), NO and SOD of the treatment groups significantly increased compared with Mod (P〈0.01). MDA and ET decreased (P〈0.01). MDA of Com decreased compared with Ben (P〈0.05). SOD of Com were increased compared with Irb (P〈0.05). Comparead with Mod, the Ang Ⅱ of Ben and Com groups significantly decreased (P〈0.01). However, the Irb increased. Conclusion:There is a synergistic effect on blood vessel endothelial function of CHF rats by combined use of benazepril and Irbesartan.
