新麦草幼胚愈伤组织诱导

Callus Induction of Immature Embryo of Psathyrostachys juncea

以山丹新麦草和新麦草新品系的幼胚为外植体,MS为基本培养基,研究不同胚龄、不同激素配比及低温预处理对愈伤组织诱导的影响。结果表明,两种材料最佳胚龄均为14d左右,幼胚大小约为1mm;激素最适配比均为2,4-D 2 mg/L+6-BA 0.2 mg/L;低温预处理最佳时间为1d。

Immature embryos of Psathyrostachys Juncea （Fisch.） Nevski cv. Shandan and a new line was used as tissue culture material. Immature embryos were cultured on MS culture medium . The effects of embryo age, exogenous hormones and the treatment with low temperature on callus induction of Psathyrostachysjuncea were studied. The results showed that the optimum immature embryo age of Psathyrostachysjuncea （Fisch） Nevski cv. Shandan and the new line were about 14d,the optimum size of immature embryo was lmm;The optimum concentration of exogenous hormones were 2,4-D 2 mg/L ＋ 6-BA 0.2 mg/L;The optimum treatment time at low temperature was ld.
