Graves病患者甲状腺肿组织中原癌基因h-ras和c—myc的表达研究

The expression of proto-oncogenes h-ras and c-myc in patients with Graves' disease

摘要 目的以β—actin为内参照物,研究原癌基因h-ras和c—myc在Graves病(GD)患者甲状腺肿组织中的表达。方法Trizol法提取26例GD和31例正常甲状腺组织标本中的mRNA,逆转录-PCR法获取cDNA,多重PCR法分别同时扩增h-ras和p—actin或c—myc和p—actin,扩增产物行1.5%琼脂糖凝胶电泳,用GelBase/GelBiot—Pro软件分析获得各标本的h-ras或c—myc和B—actin的密度值。结果h-ras在GD组中的表达较正常组有增高趋势(P=0.082),两组h-ras/β-actin的比值分别为(x±s):0.457±0.102和0.413±0.051。c—myc在GD组的表达显著高于正常组(P〈0.01),两组c—myc/β-actin的比值分别为(x±s):0.599±0.080和0.367±0.044。在GD组和正常组中h—ras和c—myc的表达均无显著相关关系(P〉0.05)。结论h-ras和c—myc在GD患者甲状腺肿组织中的表达均有不同程度的增高且两者间无显著相关关系。 Objective The expressions of proto-oncogene h-ras and c-myc were studied in patients with Graves' disease(GD) ,while β-actin was introduced in multiple PCR as the internal reference. Methods Total RNA from thyroid tissue samples,including 26 GD and 31 normal, were extracted by Trizol method and then the cDNA were obtained by RT-PCR, respectively, H-ras or c-myc was amplified with β-actin simultaneously in a multiple-PCR. The co-amplification products were separated on 1.5% agarose gel and then analyzed by GelBase/blot-Pro software, The ratio of h-ras/β-actin or c-myc/β-actin was calculated. Results In GD group and normal group the ratio of h-ras/β-actin was 0.457 ±0. 102 and 0,413 ±0.051respectively, There was an up-regulated expression tendency of h-ras in GD group (P = 0.082). The ratio of c-myc/β-actin was 0.599±0.080 in GD group and 0. 367 ± 0. 044 in normal group, respectively. The expression of c- myc in GD group was significantly increased compared to that in the normal group (P 〈0.01 ). There was no correlation between the expression of h-ras and that of c-myc in GD group, as well as in normal group ( P 〉 0. 05), Conclusions Up-regulated expression of h-ras and c-myc were observed to a certain extent, and there was no correlation between them in GD or normal group
