复健片对脑梗死大鼠运动功能和脑组织勿动蛋白-A表达的影响 被引量:7

Effect of Chinese Herb on the Motor Skill and Expression of Nogo-A in Rats with Cerebral Infarction

摘要 目的观察中药复健片对脑梗死大鼠运动功能和脑组织勿动蛋白-A(Nogo-A)表达的影响,探讨其促进神经发生的作用机制。方法60只Wistar大鼠随机分为药物组、模型组、假手术组,每组20只。用电凝法制备大脑中动脉闭塞大鼠模型,于造模成功后6h药物组灌胃给予复健片水溶液9g/kg,余2组分别灌胃给予同等量0.9%氯化钠溶液,1次/d,共2周。用横木行走实验评定运动功能;用原位杂交法观察模型大鼠梗死周边区脑组织Nogo-A的表达,并对切片进行图像分析,测定染色平均灰度。结果给药2周后3组大鼠神经功能评分两两比较,差异有统计学意义(P<0.01);药物组大鼠脑组织Nogo-A表达与模型组比较差异有统计学意义(P<0.01)。结论复健片可抑制Nogo-A的表达,从而促进神经发生,改善脑梗死大鼠运动功能。 Objective To study the effect of complex Fujian tablets ( Chinese herb) on the the motor skill and expression of nogo - A in rats with cerebral infarction and to find out its action mechanism of promoting neuragenesis. Methods Sixty rats were randomly divided into drug group ( n = 20), model group ( n = 20 ) and sham - operation group ( n = 20 ). The rat models with middle cerebral artery occlusion (MCAO) were successfully established by the eleetroeoagulation. Six hours later, the drug group were given an oral 9 g/kg dose of Fujian tablet solution, and the rest groups were given the same dose of sodium chloride solution (0 9% ) once a day for two weeks. The motor skill of the rats was determined by beam walking test. The expression of nogo - A in the region around the infarction was observed by in - situ hybridization method and the sections were subjeeted to image analysis to determine mean staining gray. Results 56 rats were involved in the result analysis. After two weeks of administration, it was noted that the score of beam walking test was more significant in the drug group than in the model group ( P 〈0.01 ) ; The expression of nogo - A in the region around infarction was more signifieant in the drug group than in the model group ( P 〈0. 01 ). Conclusion Fujian tablets can predominantly inhibit the expression of nago - A in MCAO rats and improve the neurogenesis and the motor skill of MCAO rats.
