摘要 目的探讨哮喘模型大鼠黏附分子(CD11b)、中性粒细胞弹性蛋白酶(NE)和转化生长因子β1(TGF-β1)的表达和中性粒细胞(PMN)在哮喘大鼠模型中的作用及其可能的机制。方法复制哮喘大鼠模型,随机分成哮喘组和对照组,分离纯化血PMN;采用流式细胞术测定血PMNCD11b表达,ELISA法测定NE蛋白,免疫组化法检测TGF-β1表达;对支气管肺泡灌洗液(BALF)行细胞计数。结果哮喘组大鼠血PMNCD11b、PMN和BALF中NE和TGF-β1的水平分别为(6.36±0.53)MFI,(139.33±10.72)pg/ml,(99.67±11.10)pg/ml,(0.136±0.010)OD显著高于对照组(2.65±0.45)MFI,(75.33±8.54)pg/ml,(64.89±6.75)pg/ml,(0.073±0.008)OD(均P<0.01)。哮喘组BALF中细胞总数、PMN和嗜酸性粒细胞计数分别为(6.32±0.53)×109/L-1,(13.56±2.88),(6.00±1.41)Cell/HP显著高于对照组(6.32±0.53)×109/L-1,(13.56±2.88),(6.00±1.41)Cell/HP(均P<0.01)。结论PMN、CD11b、NE和TGF-β1在哮喘时表达增加,其可能参与了哮喘的炎症过程。哮喘时PMN处于激活状态,其合成CD11b和NE的功能增强。 Objective To observe expression of adhesion molecule CD11b, neutrophil elastase and transforming growth factor-beta 1 (TGF-β1) in rat asthma model. Methods The rat asthma model was established. Blood polymorphonuclear leukocytes (PMN) were isolated and purified. CD11b was detected by flow cytometry, neutrophil elastase (NE) was detected using ELISA method, and TGF-β1 was detected by immunohistochemistry techniques. The total cell numbers and differential cell numbers in bronchoalveolar lavage fluid (BALF) were counted. The results were compared between asthma and control groups. Results Levels of CD1 lb, neutrophil elastase of PMN and BALF, TGF- 13 1 expressions in asthma group [(6.36±0.53)MFI,(139.33±10.72 )pg/ml and (99.67±11.10)pg/ml and(0.136±0.010)OD, respectively]were significantly increased than those in control group [(2.65±0.45)MFI,(75.33±8.54)pg/ml and(64.89±6.75)pg/ml,(0.073±0.008)OD, respectively](P〈0.01). Total cellular score, percentage of PMN and eosinophil in BALF of asthma group[(6.32±0.53)×10^9L^-1, 13.56±2.88,(6.00±1.41)Cell/HP, respectively]were significantly higher than those in control group [(2.78±0.36)×10^9L^-1, 5.33±1.66,(1.00±0.87)Cell/HP, respectively](P〈0.01). Conclusion Expressions of CD11b, neutrophil elastase, TGF-β1 and the count of PMN are significantly increased in asthmatic rats, which indicate that those factors may be involved in pathogenesis of asthma in rats.
