人脑胶质瘤发病分子机制及其临床应用 被引量:5

Research on molecular mechanism of human glioma and its clinical application

摘要 人脑胶质瘤是颅内最常见肿瘤,是危害人类健康和生活质量最严重的疾病之一。目前,对胶质瘤综合性治疗效果并不理想,阐明胶质瘤发病机制将为人类战胜这类疾病带来新的突破。胶质瘤发病机制是多因素作用的结果。在基因层面,Olig、p53、VEGF、PDGF、Ki-67、PCNA等基因在胶质瘤发生中起着关键作用;随着比较基因组杂交等检测方法的广泛应用,已明确胶质瘤相关染色体层面的异常变化涉及到4p、6p、9p、10q、11q、14q、14q和17p,其中1p、19q染色体的缺失与少枝胶质细胞瘤患者对放疗、化疗的敏感性和临床预后有比较明确的关系,并已指导临床工作;信号转导通路层面,胶质瘤发生中涉及到p53/MDM2/p14、p21,EGFR/PTEN/PI3K/Akt,Rb-E2F/CDK4、6/P16-cyclinD等3条信号通路变化。现已普遍认识到,应用"组学"技术和系统性疾病的观念来研究胶质瘤的发生和各种调节通路变化的关系,可以筛选出大量潜在的新型相关靶点,从而促进胶质瘤治疗学的发展进程。
