摘要
①目的探讨HP导致胃癌发生的分子生物学机制。②方法应用免疫组化SP法,检测各种基因编码蛋白在HP相关性胃炎和HP(+)胃癌组织、HP(+)胃癌与HP(-)胃癌阳性表达,比较采用秩和检验,各种基因编码蛋白间相关性采用Spearman秩相关检验。③结果HP(+)胃癌NF-κB阳性表达明显高于HP相关性胃炎,HP(+)组胃癌NF-κB及其靶基因阳性表达指数明显高于HP(-)组,差异均有显著性(P<0.05);NF-κB分别与COX-2、MMP-9及Bcl-2密切相关。④结论HP感染与胃癌的发生密切相关,NF-κB是HP相关性胃炎向胃癌转变过程中的重要因子。激活后的NF-κB可启动相应靶基因的转录和表达,参与细胞增殖与凋亡的调控及细胞转化,在胃癌发生中发挥作用。
Objective To explore the molecular biological mechanism of gastric carcinoma conduced by helieobaeter pylori (Hp). Methods The positive expressions of genes coding proteins were examined with immunohistoehemistry streptavidin - peroxidase method in gastritis infected by HP and gastric carcinoma, compared with rank sum test. Spearman rank test was used to cheek relations of these parameters. Results The positive expression rate of NF - κB in gastric carcinoma infected with Hp was higher than that in Hp associated gastritis. The positive expression rates of NF - κB and its target genes in gastric carcinoma infected with Hp were higher than those in Hp negative gastric carcinoma, and there was significant difference( P 〈 0.05 ). The expression of NF - κB was close correlated with its target genes COX - 2, MMP - 9 and Bel - 2 ( P 〈 0.05). Conclusions Hp infection was dose correlated with gastric carcinoma, NF-κB was an important factor in the, process of Hp associated gastritis changing into gastric carcinoma. Activated NF-κB could promote the target genes transcription and expression,and regulate the proliferation ,apoptosis and transformation. NF - κB played an important role in the occurrence of gastric carcinoma.
