摘要
目的观察山葡萄根提取物Vain4体外对炎性细胞功能的影响,探讨其抗炎作用机理。方法Boyden小室法测定大鼠外周血中性粒细胞趋化活性,鲁米诺发光法测定大鼠腹腔多核性白细胞超氧阴离子生成,MTT法测定人外周血粒细胞与关节滑膜细胞粘附功能和以十性红染色法观察Vam4对大鼠腹腔肥大细胞脱颗粒功能的影响。结果Vam4减少炎性三肽诱导的白细胞趋化活性,降低PMA诱发的多核性白细胞超氧阴离子生成,抑制TNFα诱导的粒细胞与滑膜细胞粘附和Compound 48/80诱发的肥大细胞脱颗粒功能。结论Vam4具有抗炎作用,其作用机理与抑制多种炎性细胞功能有关。
OBJECTIVE To investigate the inhibitory effects of Vam4, a dimer of diphenel ethylene extracted from Vitis Amurensis Rupr. on inflammatory cells. METHODS investigated the inhibitory effects of Vam4 on several pre-inflammatory models such as breath burst by chemiluminescence(CL) assay, to investigate hPMNs adherent to human synovium cells (HSC) by MTT method, chemotaxis of PBMLs by Boyden chamber method and granule release from rat peritoneal mastocytes staining with neutrol red. RESULT Vam4 presented obvious inhibitory effects on chemotaxis, breath burst, adhesion and degranulation induced by stimuli. CONCLUSION Vam4 indicated the inhibitory effects on inflammation in vitro.
