高效液相色谱法测定人血浆中头孢丙烯的浓度

Determination of cefprozil in human plasma with HPLC

目的建立一种简单的测定人血浆中头孢丙烯浓度的高效液相色谱法。方法分析柱：岛津AGT Venusil MP-C18（150mm×4．6mm，51μm）．柱温：40℃，流动相为甲醇：50mm磷酸二氢钾缓冲液（磷酸调pH=4．0）=18：82，流速：1．1mL·min^-1，检测波长：264nm。结果头孢丙烯在0．09—24．00Lg·mL^-1检测浓度范围内呈良好线性关系（r=0．9998，n=9），最低定量限为0．09μg·mL^-1，低、中、高3种浓度的日内和日间RSD〈8％。结论该方法操作简便、灵敏、准确，适用于头孢丙烯的血药浓度监测及其药动学研究。

OBJECTIVE To establish a high performance liquid chromatographic （HPLC）method for the deterruination of cefprozil in human plasma. METHODS Analytical column was AGT Venusil MP-C18 （ 150mm × 4.6mm,5 μm） , The mobile phase consisted of methanol ：50mmol · L^- 1 potassium dihydrogen phosphate （ adjusting to pH4.0 with phosphoric acid）（18： 82,V/V）with a flow rate of 1. 1mL · min^-1 ,the detection was performed at UV 264nm. RESULTS The calibration curve was linear limit of qualification was 0.09 to 24.0μg · mL^- 1 （ r = 0. 9998, n =9）. The lower limit of qualification was 0. 09μg · mL^-1. The intra-day RSD were less than 8%. CONCLUSION The method is simple, sensitive, accurate and suitable for determination of cefprozil in human plasma and pharmacokinetic study.
