摘要
用外科方法从临床确诊的细粒棘球蚴病人肝脏取出包囊,以生发层为培养材料,采用RPMI1640、199、改良伊格水解乳蛋白培养基,用胶原蛋白包被和不包被两种培养瓶,对细粒棘球蚴生发层细胞进行初培养,并进行了对比观察。结果显示,以胶原蛋白作为支撑条件,应用改良伊格水解乳蛋白培养基培养生发层细胞的效果优于其他,并已传7代,原代培养时间需要28~45d才能传代,3代以内细胞为多形态性。
The germinal cell of larval Echinococcus granulosus which excised surgically from a patient with hydatid disease has been successfully cultured in vitro. The efficiencies of culture medium RPMI 1640,199 and Eagles′MEM lactalbumin hydrolysate as well as of culture flasks with or without collagen coated were compared. The results indicated that the Eagles′MEM lactalbumin hydrolysate and collagen coated flasks were beneficial for the germinal cell of larval Echinococcus granulosus to growth. The cells had been cultured at passage 7th. The subculturing of the cells must maintain for 28 to 45 days. The cultured cell at passage 3rd revealed a variety of morphogenesis.
