高效液相色谱法测定曲安奈德氯霉素溶液的含量 被引量:2

摘要 目的建立用高效液相色谱法测定曲安奈德氯霉素溶液含量的方法。方法用十八烷基硅烷键合硅胶为填充剂,0.1%十二烷基硫酸钠(用稀硫酸调pH值至3.0)-乙腈(55∶45)为流动相,检测波长为0~12min与22~30min时为240nm,12~22min时为220nm。结果氯霉素质量浓度在0.06~0.6mg/mL、盐酸萘甲唑林质量浓度在2~20μg/mL、醋酸曲安奈德质量浓度在4~40μg/mL范围内与峰面积线性关系良好。回归方程醋酸曲安奈德为Y1=34397.2C1-19270.8(r1=0.99996),氯霉素为Y2=12109437.6C2+37543.8(r2=0.99997),盐酸萘甲唑林为Y3=321438.5C3-15109.6(r3=0.99997)。结论方法灵敏、准确、简单、快速,可用于曲安奈德氯霉素溶液的含量测定。 Objective To develop a HPLC method for quantitative determination of triamcinolone acetonide acetate and chloramphenicol solution. Methods The instruments.reagents and condition adopted in the analysis were as follows:a Shimapak- C18,UV detector of(0 - 12 min and 22- 30 min)240 nm, (12- 22 min)220 nm. The mobile phase of 0. 1% sodium lauryl sulfate(adjusted pH to 3.0 with H2SO4)-acetonitrile(55 : 45). Results The linear range of chloramphenicol was 0. 06 -0. 6 mg/mL,the linear regression equation was Y= 12 109 437.6C + 37 543.8, r = 0, 999 97 ;The linear range of triamcinolone acetonide acetate was 4 - 40 μg/mL, the linear regression equation was Y = 34 397.2 C - 19 270. 80, r = 0. 999 96. The linear range of naphazoline hydrochloride was 2 - 20 μg/mL,the linear regression equation was Y = 321 438.5 C- 15 109.6, r = 0. 999 97( n = 8). Conclusion The method is sensitive,accurate,precise and reliable for the determination of Triamcinolone Acetonide Acetate and Chloramphenical solution.
