运用溶度参数原理研究HPLC图谱特征峰位位移规律

摘要 目的探讨不同梯度洗脱条件下HPLC图谱特征峰位位移变化的规律,快速建立优化的图谱条件。方法采用HPLC法,以大黄为模型药材,比较两个不同梯度洗脱条件下特征峰的总洗脱液溶度参数。色谱柱:Alltech Apollo C18柱(250 mm×4.6 mm,5μm);流动相:乙腈-水-1%冰醋酸,梯度洗脱;检测波长:254 nm;柱温:40℃;进样体积10μL。结果大黄13个相匹配的特征峰,在初始程序和优化程序下,各自的总洗脱液溶度参数基本相等,其中1号匹配峰的偏差最大,为-12.05%,4号匹配峰的偏差最小,为-0.388%。结论HPLC图谱在不同梯度洗脱条件下特征峰位位移遵循总洗脱液溶度参数相等的规律。 Objective To investigate the displacement rule of characteristic peaks in HPLC print with different gradient elution programs and establish an optimized HPLC print. Methods The mathematic expression of characteristic peak displacement for HPLC were established in a theory, which was validated by using drug Radix et Rhizoma Rheio compared with the total solubility parameters of mobile phase of the characteristic peaks under bilateral different gradient clution programs, with an Alltech Apollo C18 as column (250 mm×4.6 mm,5μm), Acetone- Water-1% HAc as mobile phase, gradient elution, flow rate 1.0 mL/min, the detection wavelength 254 nm,the temperature 40℃, and the injecting volume 10 μL every time. Result There were 13 characteristic peaks of rhubarb and the each of the total solubility parameters of total mobile phase respectively was equal in the initial gradient elution or the optimum gradient clution program, in which, the maximum deviation for peak 1 was -12.05% and the minimal deviation for peak 4 -0.3881%. Conclusion The print displace of characteristic peaks follows the rule which is that the total solubility parameters of mobile phase arc equal in any different gradient elution programs.
