补肾增效液对辐射损伤小鼠脾组织p53 mRNA及Caspase-3蛋白表达的影响 被引量:8

摘要 目的:研究以补肾填精为主的补肾增效液对辐射损伤小鼠p53 mRNA及Caspase-3蛋白表达的影响。方法:设正常对照组、模型组、中药高、中、低剂量组和阳性药组,共灌胃8天。灌胃7天后,除正常对照组外,其余各组以^(60)Coγ射线照射小鼠造成辐射损伤。造模1天后取脾检测。p53 mRNA表达的检测采用原位杂交法;Caspase-3蛋白表达检测采用免疫组化法。结果:运用病理图像分析系统分析,各用药组均能上调辐射后小鼠脾组织p53 mRNA的表达,且与模型组比较,差异具有非常统计学意义(P<0.01);各用药组能下调Caspase-3蛋白表达,其中,阳性药组、高剂量组和中剂量组与模型组比较,差异具有非常统计学意义(P<0.01)。结论:补肾增效液通过调控与DNA损伤密切相关的p53基因及相关Caspase-3蛋白的表达而促进辐射损伤的修复。 Objective: To discuss the effect of BushenZengxiao Fluid based on the principle of tonifying kidney essence mainly on expression of p53 mRNA, Caspase-3 protein in spleen tissue of mice with radiation damage. Methods: The mice were divided into the control group,pathological model group,large, middle and small dosage group,positive medicine group randomly. Each was given treatment eight days by gastrogavage. After seven days of treatment, all the mice except that of the control group were exposed to the ^60Coγ ray to make the radiation damage model. One day later each spleen tissue was taken out to examine the expression of p53 mRNA by hybridization method and that of the Caspase-3 protein by immunohistochemical method. Results: Analysis through the pathological image analysis system shows that each medication group can increase the expression of p53 mRNA ,besides, they show a significant difference compared with the model group ( P〈0.01 ); each medication group can decrease the expression of the Caspase-3 protein, moreover, large and middle dosage group show a significant difference compared with the model group ( P〈0.01 ).Conclusion: BushenZengxiao Fluid can promote the repair after radiation damage by regulating and controlling the expression of p53 mRNA and that of caspase-3 protein which are closely related to the damage of the DNA.
