通络救脑口服液对老年性痴呆模型大鼠海马区生长抑素、胆碱乙酰化酶表达的影响 被引量:4

摘要 目的:通过研究通络救脑口服液对老年性痴呆(Alzheimer's disease,AD)模型大鼠大脑海马区生长抑素(somatostatin,SS)和胆碱乙酰化酶(choline acetyhransferase,ChAT)表达的影响,探讨通络救脑口服液抗AD的可能机制。方法:将SD大鼠随机分为空白对照组、假手术对照组、模型组、盐酸多奈哌齐对照组和通络救脑口服液高剂量组、通络救脑口服液中剂量组、通络救脑口服液低剂量组,采用海马立体定向注射凝聚态β-淀粉样蛋白_(1-40)制备AD大鼠模型。药物干预4周后,处死大鼠,取海马组织,应用免疫组化SABC方法检测SS和ChAT的蛋白表达,并通过病理图像定量分析研究。结果:与空白对照组比较,模型组SS、ChAT表达减少(P<0.05);与模型组比较,盐酸多奈哌齐组、通络救脑口服液各剂量组SS、ChAT阳性目标积分光密度均明显升高(P<0.05)。结论:AD模型大鼠大脑海马区生长抑素和胆碱乙酰化酶表达均明显降低,通络救脑口服液能够提高海马SS和ChAT蛋白的表达水平,从而发挥其抗老年性痴呆的作用。 Objective: To investigate the effect of TongluoJiu'nao oral solution on the expression of Somatostatin(SS) and Choline acetyltransferase (CHAT) in AD rats. Methods: SD rats were randomly divided intoblank control group, sham operation control group, model group, donepezil group and TLJN oral solution group(48mg·kg^-1·d^-1,24mg·kg^-1·d^-1,12mg·kg^-1·d^-1). The AD model was established by microinjection of β -amyloid protein ( β -ap)1-40 into the bilateral hippocampus. After being treated for 4 weeks, rats were sacrificed and the hippocampus was harvested, the expression levels of SS and ChAT were determined by SABC techniques, and the results were assessed by an image analysis system. Results: Compared with the blank control group, the protein expression of SS and ChAT was decreased significantly in model group(P〈0.05). Compared with model group, the protein expression of SS and ChAT increased obviously in donepezil group and TLJN oral solution group (P〈0.05) .Conclusion: The expression of SS and ChAT in hippocampus of AD rats decreased significantly. TNJN Oral Solution can anti-AD by regulating up the expression levels of SS and ChAT in hippocampus.
