摘要
目的观察异丙酚对内毒素致大鼠肺组织细胞因子表达的影响,探讨异丙酚抑制炎性反应的机制。方法成年大鼠20只随机分为四组(每组5只),对照组(C组)、内毒素组(L组)、内毒素+异丙酚20μg/kg/min(L+P20组)、内毒素+异丙酚40μg/kg/min(L+P40组)。在注射内毒素4小时后处死动物开胸取标本,用分光光度法测定诱生型一氧化氮合成酶(iNOS)、丙二醛(MDA)、NO2-/NO3-浓度。结果L组肺组织的因子的表达明显增高,异丙酚预处理组肺组织iNOS、MDA、NO2-/NO3-浓度较L组明显下降,且随着异丙酚剂量的增加而抑制效应增加。结论异丙酚能明显抑制内毒素诱导大鼠肺细胞因子的表达,对内毒素致肺损伤有保护作用。
Objective To study the effect of propofol on lipopolysaccharide(LPS)-induced lung injury. Methods Twenty adult Wistar rats weighing 230-260 g were randomly divided into four groups with 5 animals in each group: (1)conthol group received normal saline 2ml/h iv. (2)LPS group received LPS 5mg/kg/h after NS ivgtt for 2 hours,(3)propofol group A received LPS after propofol 20? g/kg/ rain for 2 hours;(4) propofol group A received LPS after propofol 40? g/kg/min for 2 hours; the animal were sacrified 4 hours after NS or LPS injection and chest was immediately opened, lung issue were removed and determind iNOS ,MDA , NO2-/ NO3- concentration. Results iNOS ,MDA ,NO2-/ NO3- concentration were significantly increased in LPS group as compared with those in control groups (P〈0.05) . propofol significantly reduced the iNOS ,MDA , NO2- / NO3- concentration in lung tissue. Conclusions Propofol inhibits LPS--induced iNOS ,MDA , NO2- / NO3- concentration increasing.
