胰岛素样生长因子-Ⅰ对高氧致新生鼠慢性肺疾病Ⅰ型胶原表达的影响 被引量:1

摘要 目的研究collagenI在高氧致新生鼠慢性肺疾病中的表达、作用及IGF-I对collagenI表达的影响。方法将足月新生大鼠144只随机分为高氧组和空气组,分别于实验1d,3d,7d,10d,14d和21d应用免疫组化染色分别检测collagenI和IGF-I蛋白表达,并将IGF-I和collagenI蛋白表达进行相关分析。结果高氧组在3d~10dIGF-I和colla-genI蛋白表达与空气组相比较明显降低(P<0·05);14d和21dIGF-I和collagenI蛋白表达与空气组相比较明显增加(P<0·05),IGF-I和collagenI表达呈正相关。结论collagenI与CLD时肺泡发育受阻和肺纤维化发生有关,IGF-I是影响collagenI合成的重要生长因子之一。 Objective: To investigate the expression and effect of collagen Ⅰ in full- term rats with hyperoxia- induced chronic lung diseases and the effects of insulin - like growth factor Ⅰ on the expression of collagen Ⅰ. Methods : One hundred and forty - four full -term rats are randomly exposed to hyperoxia (hyperoxia group) and to room air (room air group). CLD is induced by hyperoxia exposure. We use immunohistochemical method to detect the expressions of collagen Ⅰ and IGF - Ⅰ at 1, 3, 7, 10, 14 and 21 days after exposure. Results: The expressions of collagen Ⅰ and IGF - Ⅰ decrease significantly in hyperoxia group compared with room air group at 3d to 10d after hyperoxia exposure ( P 〈0.05 ) , otherwise increase significantly in hyperoxia group compared with room air group at 14d to 21 d after hyperoxia exposure ( P 〈 0. 05 ), The expression of IGF - Ⅰ correlates positively with that of collagen Ⅰ. Conclusion: collagen Ⅰ is involved in inhibition of alveolarization and lung fibrosis in the development of CLD, IGF - Ⅰ is one of the most important growth factors to influence the synthesis of collagen Ⅰ.
