摘要
目的研究DDPH对低氧内皮细胞条件培养液(HECCM)促肺动脉平滑肌细胞(PASMC)增生后的逆转效应及时间相关性,探讨DDPH使增生后PASMC恢复为正常收缩表型的时间。方法利用HECCM建立猪PASMC增生模型;采用细胞培养、改良的四甲基偶氮唑盐比色法(MTT)、免疫细胞化学染色测定α平滑肌肌动蛋白(α-SM-actin)等方法观察HECCM的促PASMC增生作用,及DDPH对PASMC增生后的逆转效应,并观察逆转效应与DDPH作用的时间相关性。结果(1)HECCM促PASMC增生后,PASMC由收缩表型转化为合成表型,胞质内的α-SM-actin含量下降,具有一定时间相关性。(2)在HECCM促PASMC增生后,加入DDPH可使PASMC由合成表型逆转为收缩表型,α-SM-actin含量回升,具有一定时间相关性。结论DDPH对HECCM促PASMC增生有逆转效应并呈一定时间相关性,DDPH作用一定时间后PASMC表型可基本逆转恢复至正常。
Objective To study the effect of hypoxial endothelia cell conditional medium(HECCM) on the proliferation of pulmonary arterial muscle cell(PASMC). Methods MTT assay was used to test the proliferation,immuno - cytochemistry was used to identify the expression of α - SM - actin. Results ( 1 ) HECCM could promote the proliferation of PASMC,down - regulate their expression of α - SM - actin. (2) DDPH could up - regulate the expression of α - SM - actin in PASMC which was time - dependant. Conclusions DDPH could reverse the phenotype transformation of PASMC exposed to HECCM,the action was time -dependant. After some time DDPH could reversly transform PASMC to the normal contractile phenotype.
