谷胱甘肽硫转移酶同功酶基因在食管癌组织中的表达 被引量:5

摘要 采用RNA斑点杂交分析方法,对40例食管癌组织和相对应的正常粘膜组织中谷胱甘肽硫转移酶(GlutathioneStransferase,GSTs)同功酶基因GSTπ、GSTα和GSTμ基因的表达情况进行研究,结果食管癌组织中GSTπ基因表达阳性率为77.5%,在相应的正常粘膜组织中表达的阳性率为90.0%。在食管癌组织中GSTα和GSTμ基因表达水平较低,表达阳性率分别为35.0%和20.0%,而正常粘膜组织中GSTα和GSTμ基因表达的阳性率分别为20%和15%,GSTs在癌组织中表达阳性率与正常粘膜组织相比,P>0.05。GSTπ基因表达阳性率明显高于GSTα和GSTμ基因,P<0.05。结果提示:GSTs同功酶基因在食管癌发生发展中基因转录水平增高,GSTπ是食管癌中GSTs同功酶存在的主要形式。 To further characterize the alteratons of GSTs gene expression during the progression of esophageal carcinoma,RNA levels of GST α,μ and π in 40 cases of esophageal carcinoma and their corresponding normal tissues were examined with Dig labeled GST cDNA probe and dot hybridization.The hybridization results was that in 77.5% cancerous tissues and in 90.0% corresponding normal tissues,an increased RNA expression of GST π was found.The positive expression rates of GST α and GST μ in the cancerous tissues were 35.0% and 20%,but in the corrresponding normal tissues were 20% and 15%,respectively.The positive expressions of GST α and GST μ were significantly lower than GST π,( P <0.05).In some cases,no expression of GSTs gene was detected in the cancerous tissues.However,in the corresponding normal tissue increased expression of GSTs gene was observed.This study suggested that GSTs gene could play an important role in development of esophageal carcinoma,and that GST π was the major form of GSTs presented in esophageal cancer.
