An Update on Clinical Application of Sildenafil

西地那非(Sil)1998年问世以来,从基础到临床进行了广泛的研究,成果丰硕。循证医学资料表明,Sil治疗勃起功能障碍(ED)具有良好的安全性、耐受性、有效性。对不同病因和多种伴发疾病以及并用除硝酸酯类外药物的ED患者均有显著疗效;还认为Sil长期服用防治ED是可行的。Sil提高cGMP水平,在细胞信号转导系统中,发挥重要作用,具有较广泛的生物活性和药理作用。因此,Sil治疗非ED疾病的信息受到关注,本文作了重点论述。

Since the advent of sildenafil （Sil） in 1998, the progress on experiments in vitro and clinical trials has been remarkable. Evidence-based medicine information has confirmed that the use of Sil for erectile dysfunction （ED） is safe, tolerable and effective, irrespective of varying etiologies and diverse comorbidities, or receiving concurrent combinations of other drugs but nitrates, and the long-term use of Sil for ED is also feasible. Owing to the key role of cGMP in the cell signal transduction system, more biological activities and pharmacological actions of Sil have been discovered by increasing the level of cGMP. The new and emerging area of information on Sil for non-ED is highlighted in this review.
