摘要
目的建立反相高效液相色谱法同时测定儿童血清中维生素A和维生素E的方法。方法取儿童血清50出,加入500μL无醛乙醇,振荡,加入正己烷萃取,将正己烷用氮气吹干后用流动相溶解。色谱条件:Zobax SB-c18 2.1×150mm(5μm);流动相:甲醇:水(96:4),检测波长:维生素A325nm;维生素E294nm。结果维生素A在50~500μg/L浓度范围内呈线性,r=0.9996,维生素E在0.5~10μg/L浓度范围内呈线性关系,r=0.9997;维生素A和维生素E的平均回收率分别为98.18%(RSD=3.21%)和98.12%(RSD=3.41%)。结论本方法检测儿童血清中维生素A和E的浓度,方法快速、准确,样品处理简便,需要的血清量少。
Objective To establish a method to simultaneously determinate the concentration of retinal and α - tocopherol in children serum. Methods 50μl serum was added 500μl anhydrous ethanol and 1000μl hexane, the mixture was votaxed and centrifuged, then the hexane was dried by N2 gas, the vitamin then resolved with mobile phase. The HPLC system was consisted of Zobax SB -C18 2.1 × 150mm(5μm) ,mobile phase was methanol:water (96 +4), detected wavelength was 325nm for retinol, 294nm for α - toeopherol. Results The linear range of determination of retinol and α - tocopherol were 50 - 500μg/L ( r = 0. 9996 ) and 0.5 - 10μL ( r = 0.9997) respectively. The piked recoveries were98.18% (RSD=3.21%) for retinol and 98.12% RSD=3.41%)forct-toeopherol. Conclusions The method was rapld, accurate, simply operating and neededa little of serum.
