摘要
以秦艽叶片为外植体,在MS培养基上附加不同种类和浓度的激素诱导愈伤组织,并对诱导出的愈伤组织进行继代增殖培养研究.结果表明:秦艽叶片在附加2,4-D 2.0 mg/L+6-BA 0.5 mg/L的MS培养基上诱导率最高,可达95.83%;继代增殖培养,蔗糖为最佳碳源,生长调节剂以2,4-D 0.1 mg/L配合KT 1.5 mg/L、B5培养基为基本培养基对秦艽愈伤组织增殖最有利.
The leaves of Gentiana macrophylla Pall. as explants, MS medium added different kinds of hormone and proportions was used for inducing the callus of Gentiana macrophylla Pall. , and how to let the callus grow rapidly was studied. The results showed the optimal medium for callus induction was the MS medium with 2,4-D 2. 0 mg/L+6-BA 0. 5 mg/L added, and its inductivity rate amounted to 95. 83%; in the course of proliferation culturing, the optimum carbohydrate was sucrose, 2,4-D 0. 1 mg/L+KT 1.5 mg/L, B5 medium as the basal medium were benefit for callus proliferation.
