付里埃变换红外测定有不同阴离子结合和通过时膜带Ⅲ蛋白的构象变化

摘要 用付里埃变换红外(FTIR)技术研究了不同阴离子结合和通过时,脂蛋白体膜带血蛋白二级结构的变化.发现:有Cl-、NO3、SO42-结合和通过时,膜带血蛋白α螺旋均为升高趋势,其中Cl-结合和通过时,带Ⅲ蛋白α螺旋增加较少;NO3-、SO4-结合和通过时,带Ⅲ蛋白α螺旋增加较多.阴离子结合和通过时,带Ⅲ蛋白α螺旋的增加可能与带Ⅲ所形成的阴离子通道扩张有关,扩张程度的大小主要取决于阴离子的体积,基本不受其所带电荷量的影响. By using FTIR technique,it was found that the amount of α-helix of membrane Band 3 was increased when anion passing acrossed it. However,the degree of the increment was different: the increment amount of α-helix by Cl- was less than that by NO3- or SO42-, Moreover,the experimental data confirmed that the increment of α-helix depends mostly on the volume of anion, but not the charge of it. By comparison of these results,it is concluded that the increment of α-helix of Band 3 was closely related to the expansion of anion channel of membrane Band 3.
