A Simple and Rapid Method for Screening Chitin Deacetylase Producing Strains

几丁质脱乙酰酶是催化几丁质直接脱乙酰生成壳聚糖的一种酶,已报道的酶活力测定方法存在操作复杂、底物昂贵等不足,不适合用于菌种筛选。研究中建立了1种分光光度法测定几丁质脱乙酰酶酶活的新方法,该方法具有操作简便,稳定性、重现性好等特点,适合于菌种筛选工作。

Chitin deacetylase （EC 3.5.1.41） is an enzyme that catalyzes the hydrolysis of acetamine groups of N-acetyl-D-glucosamine in chitin,converting it to chitosan.Although the previous methods are very sensitive,they need costly equipments or substrates.Those methods cannot be used for screening of chitin deacetylase producing strains.In this paper we reported a new spectrophotometrie method for screening of highly efficient chitin deacetylase producing strains,and determining chitin deacetylase activity.The new method is fast,simple,and reproducible,and can therefore be used routinely for estimating chitin deacetylase activity.
