摘要
利用电镜观察检测到不正常生长的香菇菌丝中存在大量直径约30 nm的球状病毒,进一步酶解和电泳检测确定病毒的核酸为单链RNA。采用尖端生长点传代技术,经过3代传代,得到不含病毒的香菇菌株,该菌株在固体和液体培养基中的生长速度都远远高于带有病毒的菌株。脱除病毒的菌株在液体发酵中产漆酶的量也明显高于带有病毒的菌株,说明该香菇病毒对香菇的生长和产酶生理过程有明显的抑制作用。
A kind of spherical virus (diameter 30 nm) was observed in an abnormal Lentinus edodes by scanning electron microscope (SEM). Its nucleic acid type was deduced as single strand RNA by enzyme digestion and electrophoresis. A virus elimination technique was adopted in this study by subculturing using the hypha tip. A high performance strain of Lentinus edodes was breed after 3 generation subculturing which could grow much faster than the original strain both in agar plates and in liquid medium. The results of laccase production detection indicated a significant increase in the new strain suggesting that viruses could significantly inhibit the growth and enzyme production of Lentinus edodes.
