H5亚型禽流感病毒HA基因在WISH细胞中的瞬时表达

摘要 在WISH细胞中,对H5亚型禽流感病毒HA基因进行了瞬时表达的研究。首先构建了真核表达质粒pVAX-H5HA,通过酶切和PCR筛选阳性克隆后测序。然后将重组表达质粒以脂质体介导法转染WISH细胞,通过RT-PCR在转录水平检测目的基因的表达,间接免疫荧光检测目的蛋白。结果表明,HA基因在WISH细胞中进行了表达。 Transient expression of HA gene of subtype H5 avian influenza virus in WISH cell was detected in this study. First, pVAX-HSHA plasmids were constructed and initially screening by means of PCR and double digestion then further ensured by sequencing. WISH cell was transferred with pVAX-HSHA plasmids by means of lipofectamine media methods. Transient expression were evaluated by RT-PCR and immunofluorescence. The result showed that the HA gene was expressed in WISH cell.
