摘要
用显微注射法将野生型p53基因导入C57BL/6J×SJL杂交F1代受精卵的雄性原核中,以ICR品系小鼠为假孕母鼠。结果出生37只原代小鼠,其中有13只在检测前不明原因死亡,其余的24只原代小鼠中有2只基因组中整合有p53基因,整合率为8.3%。
In order to analyse the biological function of wild type p53 gene in vivo,we applied microinjection method to produce transgenic mice. The DNA fragment from pDOL-53 was injected into the male pronucleus of the zygotes from F1 hybrid mice of C57BL/6×SJL. 242 injected eggs were implanted into the oviducts of ICR foster mothers, 37 offsprings were born. Among them 13 baby mice died by unknown cause,the other 24 mice grow normally. By using PCR technique to detect the transgenes in the genomic DNA of mice, 2 transgenic founders were identified. The integration rate of exogenous genes into the genome of mice was 8. 3 %. The further work to study the function of tumor suppression of wild type p53 gene in transgenic mice is underway.
