摘要 目的实验旨在建立肺炎链球菌感染的家兔肺炎模型,对肺炎链球菌的发病机制进行初步的探讨,为药物治疗提供理论基础,为药学及药物治疗的研究建立肺炎链球菌家兔模型。方法雄性新西兰兔20只,正常对照组7只,实验组13只。在全麻下经气管插管给实验组家兔注射1 mL浓度为10^(10)cfu·mL^(-1)的肺炎链球菌。接菌后,分别于0,4,6,30,54,76 h测体温和白细胞,并于76 h后处死全部动物,计算实验过程中的死亡率。取动物肺脏进行大体标本观察,制作肺组织切片并镜下观察。将肺组织匀浆做细菌培养。正常对照组也于相应的时间点测相同的指标。结果经气管给家兔接种肺炎链球菌以后可以产生肺部感染,病理改变由轻到重表现为血管病变,急性支气管炎,局灶性支气管肺炎,实变以及肺脓肿等。大体标本观察有明显的充血及颜色变化等改变,细菌培养阳性。 OBJECTIVE To build the rabbit pneumonia model of the streptococcus pneumoniae infection,to investigate the mechanism of the streptococcus pneumoniae, to provide theories for the medicine cure. METHODS Male New Zealand rabbits were divided into control group and treatment group, 7 for the control group, and other 13 rabbits for the treatment group. For the treatment group,rabbits were injected with 0. 5 mL Streptococcus pneumoniae as the density is 10^10 cfu·mL^-1. After inocubation, the body temperature and leucocyte were determined at 0,4,6,30,54,76 h, and all the animals were killed after 76 h,motality was caculated. For each pulmonary lobe, the macroscopic aspect was observed, the lung was observed by the microscope. Bacteria were incubated and counted in a sample of lung organization homogenate. RESULTS After inoculating the streptococcus pneumoniae through trachea, the lung will be infected. Pathoiogy change were obtained, including vascular lesions, bronchiolitis, focal bronehopneumonia, confluent bronchopneumonia,and lung abscess. General observe was obtained with congest changes and color changes. The bacteria incubating was positive. CONCLUSION The rabbit streptococcus pneumoniae pneumonia experiment model was established successfully.
