顶空气相色谱法测定鸡肝散7种挥发油成分的含量 被引量:3

摘要 目的建立同时测定鸡肝散(Elsholtzia blanda)中7种挥发油成分含量的方法,并初步比较不同提取方法对挥发油组成的影响。方法采用顶空毛细管气相色谱法,INNOWAX型毛细管柱(0.53 mm×30 m,5μm),氢离子化检测器(FID),以乙酸乙酯为溶剂,采用程序升温法:初始温度60℃,以30℃·min^(-1)升至90℃,再以5℃·min^(-1)升至180℃;进样口温度200℃,检测器温度240℃。结果7种成分能较好分离,其线性范围分别为α-蒎烯33~330 mg·L^(-1)(r=0.9999);β-蒎烯32~320 mg·L^(-1)(r=0.9998);1,8-桉叶醚111~1110mg·L^(-1)(r=0.9998);樟脑238~2380 mg·L^(-1)(r=0.9996);芳樟醇140~1400 mg·L^(-1)(r=0.9997);乙酸龙脑酯65~650 mg·L^(-1)(r=0.9995);龙脑53~530 mg·L^(-1)(r=0.9995)。该法的平均回收率分别为96.6%,97.0%,97.8%,99.9%,96.5%,97.4%,97.1%。结论该法简便、准确、灵敏,可为该药材的质量控制和进一步研究开发提供科学依据。 OBJECTIVE To develop a quantitative method for the determination of seven different components of volatile oil in Elsholtzia blanda, and compare primarily their contents extracted with different methods. METHODS The contents of seven components were analyzed by headspace gas chromatography with INNOWAX capillary column (0. 53 mm × 30 m,5 μm). The column temperature raised by programming, the intial temperature was 60℃, and was raised to 90 ℃ at the rate of 30 ℃· min -1 for 30 min and then was raised to 180 ℃ at the rate of 5 ℃·min^ -1. The injector temperature was set at 200℃, the detector temperature of FID was at 240 ℃. RESULTS Seven components were completely separated. The calibration curves of α-pinene,β-pinene, 1,8-cineol, camphor, linalool, bornyl acetate, borneol, were linear within the range of 33 - 330, 32 - 320, 111 - 111 0,238 - 2 38 0, 140 - 1 400, 65 - 650, 53 - 530 mg · L^-1, the con'elation coefficient were 0. 999 9, 0. 999 8, 0. 999 8, 0. 999 6, 0. 999 7, 0. 999 5, 0. 999 5, and their average recoveries (n=9) were 96.6% ,97.0% ,97.8% ,99. 9% ,96.5% ,97.4% and 97. 1%, respectively. CONCLUSION The method is simple, accurate and sensitive, and can be used for quality control of E. blanda and its further reseach.
