摘要
利用乳酸菌筛选的富集培养法,结合碳酸钙-溴甲酚紫平板,琥珀酸平板和乳酸平板以及摇瓶培养方法,从乳制品和蔬菜汁中筛选到一株以蔗糖为碳源产L-乳酸较高的乳酸菌。以此菌株为出发菌株,通过紫外诱变、60Co诱变处理,进行L-乳酸高产菌的选育,得到一株产L-乳酸量高且遗传性能稳定的突变菌株,产酸量为52 g/L,比出发菌株提高1.6倍,残糖量降低了40%。
A new method is introduced that selecting lactobacillus of L-lactic acid high-yield strain with sucrose from dairy products and vegetable juice, which used enrichment medium, with calcium carbonate -bromcresol purple plate, amber acid plate, lactic acid plate and shaking tube. This strain was chosen as the parent strain to screen and breed high-yield strain of L-lactic acid for mutagenic treatments with ultraviolet radiation and ^60Coγ ray, the obtained mutant is high-yield and stable transmissibility, the acid amount is 52 g/L, increasing 1.6 times from parent strain, the residual sugar reduced 40%.
