有机磷农残检测用植物酯酶的分离纯化及酶学性质的研究 被引量:9

Study on purification and characterization of plant-esterase for detecting the residues of organophosphate pesticides

摘要 从荞麦面粉中提取得到植物酯酶,粗酶经硫酸铵分级沉淀、Q-Sepharose Fast Flow离子交换柱层析后酶活可提纯16.814倍。纯酶液经Sephadex G-200层析和HPLC分析证明为单一物质。酶作用的最适温度为45℃,最适pH值为6.5,在40℃以下、pH值6.5~8.0之间酶活较稳定。 Plant-esterase was separated from buckwheat. The enzyme was purified to 16.614 folds through ammonium sulfate precipitation, Q-Sepharose Fast Flow. The enzyme was a single matter, which examined by Sephadex G-200 column chromatography and HPLC. The Optimum temperature and pH value for plantesterase activity were 45 ℃ and 6.5, respectively. It was more stable below the temperature 40 ℃ and in the pH rang of 6.5-8.0.
