IL-2和IL-3基因共转染白血病细胞的体内外生物学特性

摘要 目的:研究白细胞介素2(IL-2)、白细胞介素3(IL-3)基因共转染瘤苗的体内外生物学特性。方法:用IL-2重组腺病毒(Ad-IL-2)、IL-3重组腺病毒(Ad-IL-3)转染FBL-3红白血病细胞株,观察其体外增殖能力及体内致瘤性的变化。结果:IL-2、IL-3共转染的FBL-3细胞1周内能较稳定地分泌IL-2和IL-3,体外增殖能力无明显变化,但体内致瘤性明显下降,IL-2、IL-3共转染的FBL-3体内致瘤性下降最为显著,肿瘤生长明显延缓,存活期显著延长。结论:IL-2,IL-3基因的转入导致FBL-3细胞体内致瘤性的下降可能是由于激发体内免疫功能所致。 Objective:To study the growth characteristics of IL 2 and IL 3 genes co transfected FBL 3 cells. Methods: The FBL 3 cells were transfected with Ad IL 2 and /or Ad IL 3, in vitro proliferation and in vivo tumorigenicity were observed. Results: High levels of secreted IL 2 and IL 3 could remain for one week after transfection, and the transfected leukeimia cells became unchanged in vitro growth and decreased tumorigenicity. The tumorigenicity decreased most significantly when FBI 3 cells were transfected with both IL 2 gene and IL 3 gene. The tumor growth was significantly delayed and survival time was significantly prolonged. Conclusion: The inhibition of tumor growth is most likely dependent on IL 2 and IL 3 induced immune response in vivo.
