摘要
应用SSR技术开发了甜樱桃(Prunus avium)指纹检索系统并进行遗传多样性分析。18对樱桃、桃和杏的引物在19份甜樱桃及2份草原樱桃(P.fruticosa)品种中共扩增出83个等位位点,每个SSR位点的等位位点数2-8个,平均4.6个,多态性信息量(PIC)变化范围为0.38-0.80,平均为0.64。7个甜樱桃品种具有特殊位点或特殊带型。利用UDP98-414、UDP98-406、UDP96-001及PMS40等4对引物开发的指纹检索系统,可以区分18个甜樱桃品种。根据遗传距离进行聚类分析,19个甜樱桃品种分成2组,甜樱桃品种间的聚类结果基本反映了供试材料之间的亲缘关系。
SSR technique was used to develop fingerprinting key and study the genetic diversity on sweet cherry. Eighteen pairs of primers cloned from cherry, peach and apricot amplified a total of 83 alleles among the 19 cuhivars of sweet cherry ( Prunus avium) and 2 cultivars of ground cherry ( P. fruticosa). The number of alleles amplified per primer pair ranged from 2 to 8, with a mean of 4. 6. The polymorphic information content values (PIC) ranged from 0. 38 to 0. 80, with a mean of 0. 64. Seven sweet cherry cuhivars showed unique bands or banding patterns. A fingerprinting key developed by 4 pairs of primers: UDP98-414, UDP98- 406, UDP96-001 and PMS40 could discriminate 18 sweet cherry euhivars. UPGMA cluster analysis of genetic distance separated the 19 euhivars of sweet cherry into two groups. The phylogeny of sweet cherry euhivars revealed the closed relationships among euhivars tested.
