摘要
在离体培养条件下,比较了秋水仙素不同浓度、不同处理时间诱导彩色马蹄莲体细胞染色体加倍的效果。结果表明,0.05%秋水仙素处理24h的诱变效果最佳,诱变率高达52%。用组织培养结合不定芽诱导技术获得了稳定的多倍体。对变异株进行细胞学观察后发现,二倍体对照2n=2x=32,变异株2n=4x=64,为四倍体,另外还发现六倍体细胞的存在。
Coloured Zantedeschia aethiopia was treated with colchicines at different concentration and time duration in vitro. The results showed: the best effect was obtained by the treatment of 0. 05 % colchicines with 24 hours, and its induction rate was 52%. Chromosome observation found that the chromosome number of treated plants was 64, and the chromosome number of the control was 32, indicating the tetraploid (2n = 4x = 64) of coloured Zantedeschia aethiopica is induced successfully.
