摘要
在pH4.41 Britton-Robinson缓冲介质中,白蛋白(BSA)与二碘荧光黄(DIIF)作用形成复合物,使最大波长377 nm的共振光散射光谱得到加强,根据其共振光散射的增强效应,可用于蛋白质的定量测定.研究了复合物反应的适宜条件,结果表明:在优化的条件下,蛋白质浓度在0.0-5.0μg/mL范围内与共振光散射强度呈良好的线性关系,方法的检出限为0.027μg/mL.用于合成样品中蛋白质的测定,获得满意结果.
A new method for the determination of proteins is developed, based on the binding reaction of Diiodofuorescein with proteins in Britton-Robinson buffer( pH4.41 ). Diiodofuorescein Combines proteins to form complexes, causing an enhancement of resonance ralyleigh scattering(RRS) intensity at maximum wavelength of 377 nm. By means of the enhancement effect, this mehtod can be applied to the determination of proteins. The suitable conditions for the complex reaction are established. The linear range of the method is 0. 0-5. 0μg/mL of protein. The detection limit is 0. 027μg/mL. The method has been applied to determination of protein in mixed samples. The result is satisfactory.
