摘要
目的观察丹红注射液和三磷酸腺苷(ATP)对兔急性心肌梗死(AMI)的治疗性血管再生作用。方法对37只日本大耳白兔行冠状动脉结扎术,将成功存活的29只AMI模型兔随机分为4组:组1(n=5只)予NS 2 mL.kg-1;组2(n=8只)予丹红注射液2 mL.kg-1;组3(n=8只)予丹红注射液1 mL.kg-1;组4(n=8只)予ATP 2 mL.kg-1;均于耳缘静脉注射,连续给药14 d后将其处死,取心脏标本,进行免疫组化染色,测定血管内皮细胞生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)和细胞分化因子34(CD34)阳性染色值。结果丹红组和ATP组的VEGF、bFGF和CD34阳性染色值明显较阴性对照组高,丹红组高于ATP组。结论丹红注射液有明显缩小心肌梗死面积、增强梗死后心肌血管再生的能力,且效果优于ATP。
AIM To investigate the therapeutic angiogenesis effect of Danhong injection on rabbit with acute myocardial infarction. METHODS After 37 female Japan-sino hybridization white rabbit had been ligated anterior descending branch of coronary artery, 29 survivors were randomly divided into 4 groups and each one was treated with a kind of different drug respectively: group 1 administered NS, 2 mL·kg^-1, iv, qd × 14 d;group 2 administered Danhong injection, 2 mL·kg^-1, iv, qd × 14 d; group 3 administered Danhong injection, 1 mL·kg^-1, iv, qd × 14 d; group 4 administered adenosine, 2 mL·kg^-1,iv,qd × 14 d. After 14 d, their hearts were removed and cut into slices that were handled with the immunity histochemistry study of vascular endothelial cell growth factor(VEGF), basic fibroblast growth factor(bFGF) and cell differentiation(CD34). RESULTS There were more expressions of VEGF and bFGF in both infarction zone and its border and the zone of non-infarction in group 2,3 and 4 than group 1 , and the same with the density of capillary. CONCLUSION Danhong injection not only increases the expression of VEGF, bFGF and the density of capillary but also decreases the area of the infarction zone after acute myocardial infarction, and its therapeutic effect is superior to ATP injection.
