层粘连蛋白在脑胶质瘤生物学行为中的双刃剑作用

摘要 层粘连蛋白是细胞外基质和血管基底膜的主要组成成分,每一个层粘连蛋白分子都是含α、β、γ链的异源三聚体。被认识的层粘连蛋白分子已超过15种,分别由α1、α2、α3、α4、α5、β1、β2、β3、γ1、γ2、γ3以不同的组合方式构成。研究表明,层粘连蛋白在脑胶质瘤中表达增高,并在其生物学行为中起双刃剑作用,一方面,层粘连蛋白能促进胶质瘤细胞黏附和迁移,促进胶质瘤脑内侵袭,另一方面,层粘连蛋白参与抑制胶质瘤颅外转移屏障的构成。 Laminin is a major component of extracellular matrix and vascular basement membranes.Each laminin molecule is a heterotrimeric glycoprotein composed of one alpha,one beta,and one gamma chain.More than fifteen laminin isoforms have been recognized,which is assembled from various combinations of 5alpha,3beta,and 3gamma chains.Studies indicated that it played a double-edged role in the biological behaviour of cerebral gliomas in which its expression is increased.On the one hand,it can enhance the adhesion and migration of glioma cells,promote the invasion of gliomas in brain tissue.On the other hand,it participates in constituting a barrier that can inhibit the metastasis of gliomas.
