摘要 2型糖尿病是一种缓慢进展性疾病,其发病中心环节是胰岛素抵抗和胰岛β细胞功能缺陷,尤其是胰岛素分泌第一时相的损害。现已明确,即使轻度的高血糖也会严重影响胰岛素的分泌并减少胰岛素原的合成,从而使代谢所需的胰岛素进一步减少。近年来研究发现,胰腺-十二指肠同源框1(PDX-1)与胰岛素的生物合成及分泌密切相关,PDX-1的基因变异与糖尿病的发生有一定的关系,提示PDX-1可能在糖尿病的发生、发展中起着重要作用,文章就此方面研究进展予以综述。 Diabetes mellitus is a slowly progressed disease with concurrent defects in insulin secretion and insulin action,especially the failure of insulin secretion in the cephalic phase.It has been demonstrated that high glucose,etven light high glucose,can gravely affect insulin secretion and reduce proinsulin synthesization,which can decrease insulin mass.Recently,pancreatic duodenal homeobox-1(PDX-1) was demonstrated to be associated with insulin biosynthesis and secretion,and the gene variations of PDX-1 are suggested to be linked to the pathogenesis of diabetes mellitus.This article reviewed the research progress in roles of PDX-1in the development of diabetes mellitus.
