摘要 利用高效液相色谱法,以甲醇和水的混合溶液作为流动相进行梯度洗脱,经过制备柱从原料中分离出人参皂苷Re和Rg_1的混合物。确定利用重结晶的方法进行人参皂苷Re和Rg_1的分离,确定了结晶溶剂为V_(乙腈):V_水=20:80的混合溶液;结晶温度为20℃、结晶时间为5h、混合物与重结晶溶剂的固液比为1:6(g:mL)的结晶条件。在上述重结晶条件下,可得到Re纯度达91%以上的白色粉末,其收率可达92%。对得到的白色粉末进行了结构分析,确定白色粉末就是人参皂苷单体Re,并且纯度在91%以上。 By the High Performance Liquid Chromatography, employed by methanol and water as mobile phase, in progress gradient elution, the compound of Ginsenoside_Re and Rg1, which was abstracted from the stuff through preparative column. So adopted the method of recrystallization to separate the Re and Rg1, and did many experiments to select the four influence factors. The volume ratio is 20:80; crystalling temperature is 20℃; crystalling time is 5 hours; solid-liquid ratio is 1:6(g: mL). Under these crystalling conditions, it was gained the white powder (Re〉91%), the yield is about 92%, Structure of the white powder was analyzod, compared with the literature in the same condition, the refined white powder is Ginsenoside_Re, and the content is above 91%.
