大蒜多糖组分A总多糖含量的分光光度法测定 被引量:10

摘要 目的对大蒜多糖组分A进行提取并测定其含量。方法采用0.5%草酸铵溶液提取、醇沉的方法分离纯化大蒜多糖组分A,以苯酚-硫酸显色法测定大蒜多糖组分A的含量。、结果在2.0—15.0μg/ml内,浓度与吸光度呈良好线性关系。回归方程:A=0.0806C-0.0219(r=0.9934,12=10),平均回收率为96.14%,RSD%为0.06(n=5)。大蒜多糖组分A的平均糖含量为77.40%,RSD%为3.74(n=5)。结论方法简便,结果准确,为今后对大蒜多糖的药理作用以及其它组分进一步研究奠定良好的基础。 Objective The polysaccharides A were extracted from garlic and the content was determined. Methods Polysaccharides A was extracted and purified through 0.5% ammonium oxalate and ethanol precipitation, the content was determined by phenol -sulfuric acid under 495 nm. Results From the range of 2.0 - 15.01μg/ml, there was an excellent linear shape between the concentration and absorbency. The regression equation : A = 0. 0806C - 0. 0219, ( r = 0. 9934, n = 10). The average recover) was 96.14% with RSD =0.06% ( n = 5). The average polysaccharides content from garlic was 77.40% ,RSD = 3.74% ( n = 5 ). Conclusion The method is simple and accurate.
