摘要
目的对黄褐毛忍冬进行鉴别、浸出物、含量测定特征研究,提高黄褐毛忍冬质量控制标准。方法采用薄层色谱法对黄褐毛忍冬中的绿原酸、咖啡酸进行鉴别;采用50%乙醇作溶剂,热浸法测定黄褐毛忍冬浸出物含量;以50%甲醇作溶剂,超声处理30min制备样品溶液,采用高效液相色谱(HPLC)法,选用Luna[5μm,C18(2),250mm×4.6mm]色谱柱,体积流量1.0ml/min,柱温30℃,检测波长327nm,流动相为乙腈-0.4%磷酸(13:87)测定黄褐毛忍冬中绿原酸含量。结果采用薄层色谱法有效鉴别黄褐毛忍冬中的绿原酸、咖啡酸;采用50%乙醇热浸能控制黄褐毛忍冬浸出物含量;HPLC法测定黄褐毛忍冬中绿原酸含量,绿原酸在10.49~125.9μg/ml范围内呈良好线性,R^2=0.9999,通过方法学考察,精密度、稳定性、重复性、回收率实验、耐用性实验RSD值均小于2.0%。结论建立了黄褐毛忍冬鉴别、浸出物、含量测定方法,能有效用于黄褐毛忍冬质量控制,为黄褐毛忍冬质量标准提高提供实验依据。
Objectlve To improve the quality control standard of Lonicerafulvotomentosa by mean of study on the identifying character and soakings character and contents determining character of Lonicera fulvotomentosa. Methods 1 ) Identifying the ehlorogenie acid and coffee acid in Lonicera fulvotomentosa by thin layer chromatography; 2) Determining the extract contents of Lonicerafulvotomentosa with 50% ethanol solvent by hot soak ;3 ) Determining the ehlorogenic acid contents in Lonicera fulvotomentosa with HPLC and operating condition as follows: 50% earbinol solvent, 30 minutes treated by ultrasonic to the samples, chromatographic column Luna(5 μm,C18 (2) ,250 mm ×4.6 mm), the volume flow was 1.0ml/min, the column temperature was 30~C, the detection wavelength was 327nm ,identifying and the mobile phase was aeetonitrile -0.4% phosphoric acid( 13:87 ). Results 1 ) The thin layer chromatography could identify the ehlorogenie acid and coffee acid in Lonicera fulvotomentosa effectively ; 2 ) The hot soaking with 50% Ethanol could control the extract contents in Lonicera fulvotomentosa ; 3 ) When the ehlorogenie acid contents was ranged 10.49 - 125.9 μg/ml, it showed good linearity, R^2 = 0. 999 9 with RSD 〈 2%. Conclusion The identifying method on Lonicera fulvotomentosa and the determining method on their extracts and the extract contents are established. These methods can effectively control the quality of Lonicera fulvotomentosa and so that they can supply the experiment basis for improving the quality standard of Lonicera fulvotomentosa.
