摘要 目的探讨开口箭皂苷体外对人神经胶质瘤U251细胞的作用和其相关机制。方法开口箭皂苷作用于U251细胞后检测其IC50值。溶血实验观察开口箭皂苷对细胞膜的影响。提取U251细胞的DNA分析条带。RT-PCR技术检测开口箭皂苷对U251细胞的bax和bc l-2基因表达的影响。利用激光共聚焦显微技术研究开口箭皂苷对U251细胞内游离钙浓度和pH值的影响。结果开口箭皂苷对U251细胞的IC50值为8.5μg/m l。不能引起肉眼可见的溶血现象。开口箭皂苷诱发了肿瘤细胞的凋亡,并引起了bax基因表达量的升高和bc l-2基因表达量的下降,并可引起U251细胞内游离钙浓度和pH值的降低。结论开口箭皂苷体外可以诱导U251细胞凋亡,其机制可能与上述实验结果有关。 Objective To investigate the underlying mechanisms of STCB on the apoptotic induction of human neuroglioma cells. Methods The inhibitory effect of STCB on U251 cell line was studied. The haemolysis of STCB was observed. The differences of express of bax and bcl - 2 genes was compared by RT - PCR, Laser confocal scanning microscopy was used to observe the effects of STCB on the intra - cellular concentration of free cytosolic calcium and the hydrogenion concentration of U251 cells. Results The 50% inhibiting concentration was 8.5μg/ml. STCB did not cause hemolytic crisis but induced the apoptosis of U251 cells with declining expression of bcl - 2, raising expression of bax and decreasing of the intra - cellular concentration of free cytosolic calcium and the hydrogenion concentration of U251 cells. Conclusion The underlying mechanisms of STCB on the apoptotic induction of U251 cells may be associated with the results mentioned above.
