复方清脂颗粒对大鼠的肝毒性实验研究

摘要 目的研究复方清脂颗粒对大鼠肝脏的毒性作用。方法大鼠连续灌胃给予复方清脂颗粒生药52,26和13 g/kg连续8周,同时设对照组。禁食12 h后用水合氯醛麻醉动物,取肝脏称重并计算肝系数,光镜下做病理组织学检查,并测定血清中AST,ALT,ALP和肝组织中SOD和MDA。结果复方清脂颗粒52 g/kg组大鼠的肝重、肝系数、肝组织中MDA量都明显高于对照组,但SOD活力与对照组之间没有明显差别,26 g/kg组肝系数和肝组织中MDA高于对照组,13 g/kg组所有指标与对照组之间均无显著差异。病理组织学检查各组均未见肝组织有明显病变。结论长期给予复方清脂颗粒52,26 g/kg,对肝脏有一定毒性但没有造成肝脏的器质性损伤,13 g/kg是无毒性作用剂量。 Objective To study the side - effects of Fufang Qingzhi (FFQZ) granules in rat liver. Methods Rats were intragastrically treated with Fufang Qingzhi granules( 52, 26, 13g/kg) once a day for 8 weeks and the control group with water. Rats were anesthesia by chloral hydrate after fasting for 12 hours. Liver weight was measured, and liver coefficient was calculated. Histopathological examination for the liver was also performed with light microscopt. Activities of aspartate amino transferase (AST), alanine aminotransferase (ALT) , alkaline phosphatase (ALP) in serum and activity of superoxide dismutase (SOD)and concentration of malondialdehyde(MDA) in liver tissue were determined. Results Liver weight ,liver coefficient and concentration of MDA increased significantly in rats between FFQZ group of 52g/kg and control group, but activity of SOD was no different from controls. Liver coefficient and concentration of MDA increased significantly in rats with FFQZ group of 26g/kg . No significant changes in indicators were found in the rats with FFQZ group of 13g/kg compared with control group. There were no histopathological changes of the liver in all rats. Conclusion By long -term administration, FFQZ of 52 and 26g/kg has some hepatotoxicity in rats, but no histopathological changes were observed in liver. 13g/kg is the nontoxic dose.
